Název práce: Experimentální stanvení vazebných afinit fosfoproteinů vázaných do 14-3-3 proteinů důležitých pro neurobiochemii mozku.

14-3-3 proteins play key role in the neurobiochemistry of brain and are also associated with neurodegenerative amyloid diseases such as Alzheimer and Parkinson disease. 14-3-3 proteins bind more than 850 diverse target phosphoproteins, thereby forcing conformational changes or/and stabilizing active conformations in their target proteins. The aim of this PhD project is the experimental determination of binding affinities of selected phosphorylated proteins with 14-3-3 by NMR, ITC and fluorescence titration experiments. The PhD student will prepare all proteins needed for this project including phosphorylation of the 14 3 3 target proteins and their fluorescence labeling. The binding affinities obtained from the different experimental methodologies of small phosphopeptides binding to 14-3-3 will be compared to computationally predicted binding affinities.
