Posttranscriptional regulation of gene expression is mediated by specific RNA-protein interactions which determine time and space of protein synthesis within the cell. Human Staufen 1 (STAU1) protein is an important double-stranded RNA binding protein, which regulates transport and decay of certain mRNAs in the cytoplasm, but the molecular basis of how STAU1 recognizes its targets is still unkown. We plan to study the molecular interactions between STAU1, 3’UTR mRNA sequences and long non-coding RNAs which select specific mRNAs for STAU1-mediated mRNA decay using biochemical methods and NMR spectroscopy.
