Název práce: Strukturní studie viru deformovaných křídel, jako modelového organismu ze skupiny iflavirů infikujících včelu medonosnou

One tenth of agricultural crop production depends on insect pollination that is predominantly provided by honeybees. However, the number of honeybee colonies in the US has declined from 5.5 millions in 1950 to 2.1 millions in 2010 and similar decline is estimated for the EU. It has been shown that virus infections are one of the major factors contributing to the collapse of honeybee colonies. Most of honeybee viruses belong to families iflaviridae and dicistroviridae. Currently, no structures of honeybee viruses are known. The student will use X-ray crystallography to study structures of DWV virions, empty particles and of non-structural proteins RNA-dependent-RNA-polymerase 3Dpol, protease 3C, and RNA-helicase. The student will explore possibilities to prepare large quantities of the virus including isolating the virions from infected honeybees and using baculovirus expression system to produce DWV virions in cells that are normally not permissible for DWV. Subsequently, he/she will determine crystallization conditions. He/she will use molecular replacement to solve the structures of DWV virions and empty particles. The non-structural proteins will be expressed in bacterial cells. The student will also explore the possibilities to adapt picornavirus inhibitors for the use against DWV.
