Beef extract 10 g
Peptone 10 g
Agar 20 g
Sea water* 750 ml
Tap water 250 ml
Dissolve beef extract and peptone by heating in
water, adjust pH to 7.8, boil for 10 min.
Readjust pH to 7.3 and add agar.
* Natural sea water is stored in the dark for at
least three weeks to "age". If natural sea water
is not available, use artificial sea water.
Artifical sea water:
NaCl 28.13 g
KCl 0.77 g
CaCl2 1.2 g
MgCl2 2.55 g
NaHCO3 0.11 g
MgSO4.7H2O 3.5 g
Distilled water 1000 ml
