The role of nitrate-inducible bacterial response regulator homologs in Arabidopsis thaliana

Response regulators are regulatory proteins from two-component systems common in prokaryotic cells. Recently, several response-regulator-like genes were found in Arabidopsis and other eukaryotes. Some of these genes (ARR3-ARR7) were shown to be induced b y nitrate and plant growth regulators. Given their presumed regulatory function, this makes them good candidates for early response or signal transduction proteins for nitrate. Reverse genetics can be used to assign biological function to genes of known sequences. In this study, we will use PCR to screen Arabidopsis transpozone libraries for insertions in ARR genes and their close homologues. The mutations will be isolated in homozygous state in a wild-type backround. Subsequently, we will characterize their effect on processes regulated by nitrate and growth regulators. If ARR proteins indeed serve as nitrate early response or signal transduction proteins, then these processes should display altered response to the respective chemical stimulus in plan
