The simultaneous activity of transcription and replication or DNA repair in the same genomic region can lead to genome instability and requires tight regulation. However, the involved mechanisms remain largely unknown. In this project, I aim to shed light on this area of research using the SMC5/6 complex, which seems to play a major role in bridging these competing processes. I plan to show that the DNA repair regulated by SMC5/6 leads to the inhibition of transcription to prevent genome rearrangements, which can potentially lead to cancer. Furthermore, the addressed SMC5/6-dependent transcription inhibition mechanism may also be utilized in its role in the antiviral response, making this research of utmost importance.
