ADARs converts adenosine to inosine in double-stranded RNA and this is one of the most abundant modifications found in RNA. Previously, my group has demonstrated the pivotal role of ADAR1 in innate immunity. However there is also an editing independent function of ADAR1 that is important but currently unknown. We propose to uncover this editing independent function by investigating proteins that interact with ADAR1. We have preliminary data on the interaction of ADAR1 with differential protein complexes when cells are treated with α-interferon. We will analyse this data, validate protein interactions and map which biological pathways ADAR1 has a direct influence on and whether it depends on the enzymatic activity.
It has been predicted that ADAR1 is highly modified posttranslationally and this may account for variation in its activity in different cell types. The second aim is to elucidate if these posttranslational modifications regulate ADAR1, in particular those modifications of ADAR1 that occur after type 1 interferon induction.
