Sensory organelles termed primary cilia have emerged as elaborate signaling compartments with crucial roles in both development and disease. Thus, a firm understanding of primary cilia biology is critical to comprehend the roles these organelles serve in human health. In turn, the key to understanding the structure and function of primary cilia is the identification of individual cilia components and thorough characterization of how they act together to assemble and maintain functional organelles.
The project we propose here is centered on the elucidation of new cilia-related roles and the underlying molecular mechanisms of both novel (SAMD4B) and established (TTBK2) regulators of primary cilia function. The project combines several state-of-the-art methodologies (advanced microscopy, proteomics, etc.) with cellular and animal models, hence allowing for both unbiased and hypothesis-driven experimental strategies. We are convinced the proposed research has the potential to significantly advance our insight into relevant molecular mechanisms controlling primary cilia in health and disease.
