Proposed project is established on searching for new methods suitable in metallopeotein analysis. It is based on hyphenation of chosen separation methods with selective detection of species containing metal elements. Among proposed methods, used in protein separation, we have included beside the more common gel electrophoresis in polyacrylamide gel also so far unexploited methods with high separation potential such as continuative elution gel electrophoresis and high-performance liquid chromatography on monolithic sorbents. Detection strategy lies in parallel identification of respective protein using MALDI-TOF mass spectrometry and of respective metal element of given metalloprotein by means of mass spectrometry with inductively coupled plasma ionisation. Hyphenation of separation and detection techniques will be realised in dependence of separation output, as in solution as in gel. For direct elemental analysis of gel, laser ablation will be utilised, whereas for solution analysis, it will be nebulisation techniques.
